    Flowering of kiwifruit (Actinidia deliciosa) is reduced by long photoperiods

    Mature kiwifruit (Actinidia deliciosa ‘Hayward’) vines grown under standard orchard management were exposed to 16-h photoperiods from the longest day in summer until after leaf fall in autumn. Photoperiod extension was achieved with tungsten halogen lamps that produced 2–8 ”mols m–2 s–1 photosynthetically active radiation. Long day treatments did not affect fruit dry matter or fruit weight at harvest during the growing season that the treatments were applied or during the following growing season. However, flowering was reduced by 22% during the spring following treatment application. As this reduction in flowering was not accompanied by a decrease in budbreak, the long day effect is not consistent with a delay in the onset of winter chilling. It is suggested therefore, that the observed reduction in flowering may be because of a diminution of floral evocation

A common concern is the non-attendance at lectures of unmotivated (conscript) students. Poor lecture quality, as reflected in student evaluations of teaching (SETs), is often blamed for lack of attendance and consequent poor performance. This paper presents the results of a student assessment of a macroeconomics 1 course, coupled with a self-assessment of their own input into the course. The results obtained, using econometric models, suggest that students inputs and attitudes to the course are equally, or more, important than lecture attendance itself

    The willingness to pay for dusky kob (Argyrosomus japonicus) restocking: using recreational linefishing licence fees to fund stock enhancement in South Africa

    The economic feasibility of stock enhancement of Argyrosomus japonicus in South Africa was investigated using a willingness-to-pay (WTP) survey. The pilot study provides a unique example of the use of the contingent valuation method as a valuation tool for a proposed stock enhancement programme. An increase in the cost of a recreational fishing permit is used as a potential vehicle of payment. The median value of the maximum that fishers were willing to pay for a recreational fishing permit was R155 (South African Rand) for frequent fishers and R100 for non-frequent fishers. Analysis showed that a fee of more than R100 excluded up to 50% of anglers from the fishery, but that a fee of R100 excluded only 28% of recreational anglers and would generate an additional R12 million annually from the sale of recreational fishing permits. The estimated costs of set-up and running of a stock enhancement programme are substantially lower than this, suggesting that stock enhancement may be an economically feasible management option that deserves more investigation. The WTP method itself produces robust results and is likely to be an effective tool in the management of the marine environment

    Factors enabling and constraining research in a small, research-intensive South African University

    preprintResearch is increasingly regarded a core facet of university endeavours globally, and research profiles of universities, institutes, and colleges are commonly used as one measure for ranking them. University administrations and funders would be better able to stimulate research if they had insights into context-specific, institutional constraints, and enablers. Yet, there is surprisingly little research on the determinants of research productivity amongst academics employed in the higher education sector, particularly in the global south. Barriers and enablers of research at the individual level may differ, and experience of such may vary across career stage. The objective of the research reported here was to determine what enables and motivates some academics at Rhodes University (South Africa) to do research, what problems and constraints may be making it difficult for those who may wish to do more, and how do these vary in relation to research career stage. An anonymous online survey, with follow-up focus group discussions was used to collect the data. Results show similarities to studies in developed countries, but also reveal disciplinary and career stage differences, which suggest that institutional policies need to take these factors into account when designing support or incentives

    Adaptation of Quinoa (Chenopodium quinoa Willd.) to Australian Environments

    Quinoa is being evaluated in cropping systems in many countries outside of its natural range of South America. Very few attempts have been made by farmers or researchers to grow or evaluate quinoa under Australian environments. Given the growing popularity of quinoa with consumers, new commercial opportunities for farmers and international interest in the crop, it was timely to undertake a comprehensive evaluation of the potential of quinoa in Australia. Two advanced selections and nine germplasm lines (six of Chilean and three of Bolivian origin) identified in an earlier project were tested in 23 field trials at 14 locations on mainland Australia. Targets included irrigated sites in tropical, Mediterranean, semi-arid and desert climates, and rain-fed sites of south-western Australia with a Mediterranean climate. The field experiments were either a randomised complete block design (RBCD) or a split plot/factorial design with 2–4 replicates, and a linear mixed model was used to compare the treatment lines. Seed yield of quinoa was highest when grown in winter and spring under rain-fed conditions in Geraldton, in spring and summer under irrigation at Bool Lagoon, and summer, autumn and winter under irrigation at Leeton. The highest seed yield achieved was 3 t/ha for a germplasm line from Chile, while the highest yield for a germplasm line from Bolivia was 2.6 t/ha. Advanced selections from Australia yielded well in comparison at most trial sites. Declining seed yield was associated with mean daily temperatures during seed development increasing above 17 °C, mean daily temperatures during flowering declining below 15 °C, and rainfall during seed development under rain-fed conditions falling below 50 mm. Seed produced at Bool Lagoon was the closest in colour to white quinoa imported from Peru; however, it was more than noticeably different. Seed produced at Geraldton and Leeton was significantly larger than from other field sites; however, none were larger than 2 mm in diameter as found in Royal white quinoa from Bolivia. Superior seed colour and seed size were associated with dry conditions at maturity and cool conditions during seed development, respectively. We conclude that quinoa can become a potential crop option for Australian agriculture by exploiting genetic diversity and supplementing with suitable management practices matched to agro-climatic environments. There are reasonable prospects to raise the seed yield potential in areas in all states, especially in the regions where quinoa grew well in our experiments

    Selecting Grassland Species for Saline Environments

    In Australia, around 5.7 million hectares of agricultural land are currently affected by dryland salinity or at risk from shallow water tables and this figure is expected to increase over the next 50 years (LWRA, 2001). Most improved grassland species cannot tolerate the combined effects of salt and waterlogging and, therefore, the productivity of sown grasslands in salt-affected areas is low. However, there is potential to overcome the lack of suitably adapted fodder species by introducing new, salt and waterlogging-tolerant species and by diversifying the gene pool of proven species. Potential species include exotic, naturalised and native Australian grass, legumes, herb and shrub species that are halophytes and non-halophytes. A collaborative national project in southern Australia commenced in 2004 with the objective of evaluating a range of forage species for saline environments

    In vivo CRISPRa decreases seizures and rescues cognitive deficits in a rodent model of epilepsy

    Epilepsy is a major health burden, calling for new mechanistic insights and therapies. CRISPR-mediated gene editing shows promise to cure genetic pathologies, although hitherto it has mostly been applied ex vivo. Its translational potential for treating non-genetic pathologies is still unexplored. Furthermore, neurological diseases represent an important challenge for the application of CRISPR, because of the need in many cases to manipulate gene function of neurons in situ. A variant of CRISPR, CRISPRa, offers the possibility to modulate the expression of endogenous genes by directly targeting their promoters. We asked if this strategy can effectively treat acquired focal epilepsy, focusing on ion channels because their manipulation is known be effective in changing network hyperactivity and hypersynchronziation. We applied a doxycycline-inducible CRISPRa technology to increase the expression of the potassium channel gene Kcna1 (encoding Kv1.1) in mouse hippocampal excitatory neurons. CRISPRa-mediated Kv1.1 upregulation led to a substantial decrease in neuronal excitability. Continuous video-EEG telemetry showed that AAV9-mediated delivery of CRISPRa, upon doxycycline administration, decreased spontaneous generalized tonic-clonic seizures in a model of temporal lobe epilepsy, and rescued cognitive impairment and transcriptomic alterations associated with chronic epilepsy. The focal treatment minimizes concerns about off-target effects in other organs and brain areas. This study provides the proof-of-principle for a translational CRISPR-based approach to treat neurological diseases characterized by abnormal circuit excitability
